【Abstract】 Objective To probe the effect of radix sophorae tonkinensis liquid for allergic asthma.Methods (1) Establish the guinea pig model of allergic asthma:divide 30 guinea pigs into three groups equally,then inject them in abdominal cavity by 10% albumin-egg.After two weeks,stimulate them into asthmatic condition by 2% albumin-egg in aerosol inhalation; (2) Extract the effective parts of radix sophorae tonkinensis by water.(3) Treat the guinea pigs by the extracted liquid in aerosol inhalation.Results (1) The curative effect of group A which is treated only by the extracted liquid is better than compared group C; (2) The curative effect of group B which is treated by the extracted liquid and CCBs(verapamil) is the best of the three groups.Conclusion The liquid extracted from radix sophorae tonkinensis has a good effect in allergic asthma,and the effect is better under the assistant with CCBs(calcium channel blocking agents).

过敏性哮喘是一种发作性的以细支气管广泛性痉挛为特征的呼吸系统常见疾病,对人类的健康危害很大。山豆根提取液中含有的苦参碱能明显对抗组胺等炎症介质兴奋气管平滑肌的作用,从而达到解痉、平喘的效果,这种作用在无钙离子的情况下更为明显。目前资料显示[1],苦参碱主要应用于心律失常、乙肝、肿瘤及炎症等方面,其曾应用于喘息性支气管炎和支气管哮喘的临床治疗观察,但并未在临床推广普及。本实验通过建立豚鼠过敏性哮喘模型,探讨山豆根提取液对过敏性哮喘的疗效及应用价值。
1 材料与方法
1.1 材料
1.1.1 实验动物 清洁级豚鼠30只,雌雄不限,体重400±50 g,由右江民族医学院实验动物中心提供。
1.1.2 试剂与器械 卵清蛋白(Albumin.egg,由北京索莱宝科技有限公司提供),盐酸维拉帕米注射液(规格:5mg/2ml,治疗时先用生理盐水稀释至0.5mg/ml,再按稀释浓度计算注射剂量),山豆根;402型超声雾化吸入器(上海医疗器械公司),密闭透明容器箱(容积:30cm×22cm×15cm),天平,注射器、烧杯若干。
1.2 方法
1.2.1 豚鼠过敏性哮喘模型的制备
常规饲养1周无异常后,均予以腹腔注射10%卵清蛋白生理盐水200mg/kg;2周后将豚鼠放置于密闭透明容器箱中并予以2%卵清蛋白生理盐水雾化吸入激发哮喘。哮喘发作判断依据与分度[2]:(1)轻度:点头呼吸,腹式呼吸运动增强,呼吸急促(呼吸频率≥1.2×基础频率)或叹气样深慢呼吸,烦躁不安。(2)中度:在轻度的基础上,伴有咳嗽、站立不稳、口唇紫绀,无大小便失禁。(3)重度:在轻度的基础上,伴有咳嗽、抽搐、倒地、口唇紫绀,可有大小便失禁。(4)死亡。根据上述分度标准,将各组豚鼠哮喘激发结果分类归纳如表1。
