医药学论文：清胃散中盐酸巴马汀等成分的含量测定

【摘要】 目的 比较在清胃散合煎液和分煎液中盐酸巴马汀和盐酸药根碱及芍药苷的含量。方法 采用HPLC测定在清胃散合煎液和分煎液中盐酸巴马汀和盐酸药根碱及芍药苷的含量和对常用菌种进行常规琼脂扩散管碟法定性测定。结果与结论 分煎液中盐酸巴马汀和盐酸药根碱含量均为合煎液中相应成分含量的2.73倍，而分煎液芍药苷含量为合煎液中相应成分含量的1.17倍。

【Abstract】 Objective To compare the content of palmatine hydrochloride, jatrorrhizine hydrochloride and paeoniflorin inthe mixed decoction and the separateddecoction and the antibacterialexperimentsof Qingwei San.Methods The three components was determined by HPLC and the qualitative test by the general agar of diffused cup-plate method and the quantitative assay by the test-tube method in the antibacterialexperiments Results The average content of the palmatine hydrochlorideand the jatrorrhizine hydrochloridewhich was the more 2.73times inthe separateddecoctionthan in the mixed decoction, and the paeoniflorinwas only 1.17 times in the separateddecoctionthan in the mixed decoction.Conclusion The resuls of this experiment for Qingwei San which is provided reference to clinic use.

清胃散出自《脾胃论》，由黄连、当归、生地、丹皮、升麻等5味中药组成。主治胃火牙痛。用分煎法与合煎法制备的中药复方汤剂，其有效成分含量、药理作用有何变化，一直是中医药理论和临床实践中值得探讨的课题，本论文以中药经方清胃散中的有效成分盐酸巴马汀和盐酸药根碱及芍药苷作为考察指标，比较该方用合煎法与分煎法制得的汤剂中有效成分含量，探索中药复方汤剂不同制法对其有效成分溶出量影响。

 

1 实验处方、仪器和药品与试剂

 

1.1 实验处方［1］ 黄连6g，当归6g，生地6g，丹皮9g，升麻9g。

 

1.2 仪器 美国惠普HP1100型高效液相色谱仪，HP1100/ WIND3D 化学工作站。 DAD 检测器。

 

1.3 药品与试剂 黄连为毛茛科黄连(Coptis chinensis Franch.)根茎，产地四川；当归为伞形科当归［Angelica sinensis (Oliv.) Diels］根，产地甘肃；生地为玄参科生地(Rehmannia glutinosa Libosch.)块根，产地河南；丹皮为毛茛科牡丹(Paeonia suffruticosa Andr.)根皮，产地安徽；升麻为毛茛科升麻(Cimicifuga foetidaL.)根茎，产地辽宁；经贵阳中医学院何顺志教授鉴定且符合2005版《中国药典》一部收载。盐酸巴马汀对照品（批号732－9906）,盐酸药根碱（批号733－9906）芍药苷对照品(批号110736-200422)，购于中国药品生物制品检定所。乙腈(色谱纯)，水(重蒸)，其余试剂均为分析纯。
