【Abstract】 Objective To observe the effects of adenosine(CPA)on obesity rats.Methods The rats were divided into three groups,one group were raised by normal food (group A),another were raised by high-energy food (group B)and high-energy food combined with CPA raised group(group C).The weight whole body were measured and the rate of heart/weight,fat/weight,liver/weight and kidney/weight,center vein pressure(CVP),blood pressure(BP) and Lee's index were calculated respectively.Results (1)The body weight,the rate of fat/weight,Lee's index,CVP,BP in high-energy food group were higher than those in normal groups(P< 0.01,P< 0.05),but the rate of heart/weight,liver/weight,kidney/weight were lower than those in normal groups(P<0.05).(2)Body weight,Lee's index,fat/weight,CVP,BP in high-energy food combined with CPA raised group were lower than those obesity rats group(P<0.05),but it has no significant difference compared with normal raised group(P>0.05).(3)The heart/weight,liver/weight and kidney/weight in obesity rats group were lower than those in normal raised group,high energy food combined with CPA raised group(P<0.05),but those were no significant difference compared with normal raised group(P>0.05).Conclusion Adenosine was likely to restrain the occurence of obesity rats.

腺苷(adenosine)是体内的一种重要的生物活性物质。腺苷根据受体的生物特性可分为A1、A2a、A2b、A3等3个亚型[1]。人们在白色脂肪细胞上发现A1腺苷受体。在白色脂肪细胞的培养液中分别加入A1腺苷受体特异性的激动剂CCPA和A1腺苷受体特异性的阻断药DPCPX后发现,加入CCPA的培养液中的脂肪细胞的生长受到了明显的抑制,加入DPCPX的培养液中的脂肪细胞生长良好[2]。本实验采用腺苷受体特异性的激动剂(N6-cyclopentyladenosine,CPA),旨在探讨腺苷对肥胖大鼠整体水平的影响。
1 材料与方法
1.1 材料
1.1.1 实验动物 选用出生1个月的健康Wistar大鼠30只,雌雄各半,体重(158±33)g,由华中科技大学同济医学院实验动物中心提供。
1.1.2 饲料 实验选用合成饲料[3]。普通饲料和高能饲料均由华中科技大学同济医学院实验动物中心加工而成。
1.1.3 试剂 N6-环戊腺苷购自Sigma公司,混合无机盐(NaCl 2.5g/100g饲料,KCl 0.1g/100g饲料,MgCl 0.9g/100g饲料),混合维生素(21金维他)购自杭州民生药业集团有限公司,酪蛋白(分析专用)购自天津市化学试剂公司。
1.1.4 主要仪器 BP211D电子分析天平(德国),ES-C200A天平(湘仪天平仪器厂),BL-410生物信号采集分析仪(成都泰盟科技有限公司)。
1.2 方法
1.2.1 大鼠肥胖模型的复制 健康Wistar大鼠30只,室温20℃~26℃,湿度80%~85%,通风良好、光照充足条件下,适应性喂养1周后,随机分出20只大鼠,采用高能饲料喂养[3]8周复制肥胖模型。剩下10只普通饲料喂养。
1.2.2 动物分组与处理 随机分为普通饲料与高脂饲料喂养组(10只,雌雄各半),即A组;肥胖模型组(10只,雌雄各半),即B组;肥胖模型组加CPA组(10只,雌雄各半),即C组,CPA腹腔注射,0.02mg/100g,稀释至0.1ml/100g注射(实验采用A1腺苷受体的特异性激动剂CPA来激活A1腺苷受体)。A组普通饲料喂养及B组、C组仍用高能饲养至第14周后处理动物。
