Dynamic observation on the biochemical index in the immunosuppressive mice experimentally infected with Taenia sagi-nata asiatica in Duyun City and Taenia saginata in Congjiang City.
Abstract:Objective To observe the dynamic change on the biochemical index in mice experimentally infected with Taenia saginata. Methods Mice were subcutaneously infected with oncospheres of T.saginata asiatica and Taenia saginata in Con-gjiang and every mouse was treated with dexamethasone(1mg/day)through subcutaneous injections.All the mice were dissected40,60and75days after infection,then the liver function,renal funtion and myocardial enzyme index was determined. Results Liver funtion TP、ALB、ALT、ASP changed more significantly with the increase of days aftger infection;renal funtion index BUN、CREA、UA and myocardical enzyme CK、LDH improved markedly,with the increase of parasitized days of cysticerci,a variety of biochemical in-dex of a multiple increased. Conclusion The biochemical indexes of mice such as liver and renal funtion,myocardial enzyme index etc.changed more significantly with the increase of infected days.

贵州省黔南布依族苗族自治州都匀市和黔东南苗族侗族自治州从江县是牛带绦虫病的流行区 [1] ,相关文献已证明都匀及从江牛带绦虫感染猪、牛后,生活史形态不同,并经DNA、PCR扩增试验体细胞色素氧化酶,基因片段并测序,计算遗传距离并构建系统发育树状圈,结果贵州都匀基因序列与已确认的台湾亚洲牛带绦虫(Taenia saginata asiatica)基因序列的同原性达97.44%,氨基酸序列到同源性达99.16%,而与从江县牛带绦虫区别不同的亚种 [2~5] 。为观察都匀对宿主组织器官的伤害,现以免疫抑制小鼠感染都匀亚洲带绦虫与从江牛带绦虫动态观察生化指标,以探讨感染对宿主生化指标的影响。
1 材料和方法
1.1 实验动物 昆明小鼠60只,雌、雄各半,体重18~20g(购自贵阳医学院动物中心),并随机分为2组,都匀亚洲带绦虫感染组(30只),从江牛带绦虫感染组(30只)。感染前小白鼠经粪检、驱虫,证明无囊尾蚴感染。
1.2 方法
1.2.1 实验虫体来源 虫来自贵州都匀亚洲带绦虫流行区和贵州从江牛带绦虫流行区已确诊的绦虫病患者,采用南瓜子、槟榔驱虫法,驱出完整的虫体,用生理盐水洗净,经形态学鉴定,贵州都匀牛带绦虫为亚洲带绦虫,从江牛带绦虫为传统的牛带绦虫。
1.2.2 虫卵收集 将用生理盐水洗净的成虫分别取其末端成熟孕节片10节剪碎,用无菌生理盐水漂洗,除去虫体碎片,1000r/min离心5min,弃上清液备用。
1.2.3 虫卵孵化 将0.5%次氯酸钠溶洲加入离心后的虫卵沉淀物中,室温振荡混匀,8min后吸取混匀的孵化液镜下观察,待大部分卵膜破裂六钩蚴孵出后,离心,去上清液加入10ml无菌蒸馏水混匀后,倒入培养皿中,再分别用60ml无菌蒸馏水洗净离心管后倒入培养皿中,混匀后镜下计数(约700个六钩蚴/ml)备用。
1.2.4 小鼠免疫抑制方法 从感染当天开始,每只小鼠皮下注射地塞米松1mg/d,同时在小鼠饮水中按0.8mg/ml的比例加入四环素以防止细菌感染,直至剖杀。
1.2.5 实验感染方法 用注射器吸取六钩蚴溶液,缓慢注入小鼠皮下和腹腔,每只注射1ml溶液。感染后的小鼠按组隔离喂养。
